Spin-label study of hemoglobin conformations in solution.

چکیده
There is now a great deal of physical and chemical evidence that horse and human hemoglobins undergo a change of protein conformation on oxygenation.' 9 The molecular mechanism whereby these hemoglobins show cooperative or sigmoidal oxygen binding has remained obscure, however. In order to understand this "heme-heme" interaction, one must obtain structural information on partially oxygenated hemoglobin molecules in solution. The present paper is a preliminary report of our study of oxygen-linked conformational changes in hemoglobin, using the spin-label technique. We also discuss the relation between conformational changes and allosteric interactions such as the cooperative binding of oxygen to hemoglobin. Materials and Methods.-Horse and human hemoglobins were prepared according to the method of Benesch and Benesch'0 from fresh defibrinated horse blood (W. T. Bennett Ranch Laboratory) and from samples obtained through the courtesy of Dr. Marcus Krupp, Palo Alto Medical Clinic. The spin label, N-(1-oxyl-2,2,5,5-tetramethyl-3-pyrrolidinyl)iodoacetamide (I),

Spin-labeled hemoglobin derivatives in solution and in single crystals.
The paramagnetic resonance of a spin-label attached to a protein in solution, or in a single crystal, depends on static as well as on the dynamic features of its molecular environment.' This conformation-dependent paramagnetic resonance offers the possibility of relating structural properties of proteins in single crystals to those in solutions. The present paper is a brief report of a study of...
متن کاملHeme-Spin Label Studies of Hemoglobin
In order to investigate the protein conformation in the vicinity of the heme and the spin states of the heme-iron in dissolved hemoglobins, a nitroxide spin label was covalently attached to one of the propionic acid groups of the porphyrin ring. The optical spectra of the heme-spin labeled ferrihemoglobins were identical to those of native hemoglobins indicating that the spin labeling did not a...
متن کاملIntegrated analysis of the conformation of a protein-linked spin label by crystallography, EPR and NMR spectroscopy
Long-range structural information derived from paramagnetic relaxation enhancement observed in the presence of a paramagnetic nitroxide radical is highly useful for structural characterization of globular, modular and intrinsically disordered proteins, as well as protein-protein and protein-DNA complexes. Here we characterized the conformation of a spin-label attached to the homodimeric protein...
متن کاملLocation of the allosteric site for 2,3-bisphosphoglycerate on human oxy- and deoxyhemoglobin as observed by magnetic resonance spectroscopy.
2,3-Bisphosphoglycerate, the predominant phosphorylated metaholite of the human red blood cell, binds tightly to human deoxyhemoglobin and weakly, yet significantly, to human oxyhemoglobin. To locate the binding site(s) for 2,3-bisphosphoglycerate in both oxyand deoxyhemoglobins in solution, we have measured the paramagnetic effects of hemoglobin spin-labeled with a nitroxide radical at each of...
متن کاملRosettaEPR: Rotamer Library for Spin Label Structure and Dynamics
An increasingly used parameter in structural biology is the measurement of distances between spin labels bound to a protein. One limitation to these measurements is the unknown position of the spin label relative to the protein backbone. To overcome this drawback, we introduce a rotamer library of the methanethiosulfonate spin label (MTSSL) into the protein modeling program Rosetta. Spin label ...
